Yes, there is a point of no return in Marvel's Spider-Man 2; specifically, this becomes available after "Finally Free" and is at the main story marker for the final campaign mission, "Together", where you need to "regroup at May's House" so you can plan to destroy the meteorite and defeat Venom.
Before starting "Together", you're free to explore New York and take on any extra side missions, gather collectables, and collect resources for upgrades as you please.
Once you approach May's front door and start the mission, you'll be locked in for the rest of the story. However, don't worry if you accidentally press the Triangle prompt at May's door by mistake, you'll first be shown the warning in the above image. You'll only enter the point of no return when you press the X button to accept the warning.
Afterwards, you'll be locked in between 45 minutes to an hour until the end of Spider-Man 2's story, allowing you to explore again after you've gone through the end credits and post-credits scenes.
